Comments:
Corticobasal degeneration is a neurological disease characterized by the progressive degeneration of multiple regions of the cerebral cortex and basal ganglia. Symptoms of the disease are similar to those of Parkinson's disease (PD), including poor motor coordination, hypokinesia, balance problems, abnormal muscular postures, cognitive impairment, hesitant and halting speech, and apraxia. ERK1-deficient mice were observed to demonstrate an enhanced striatum-dependent (striatum = caudate nucleus and putamen, parts of the basal ganglia) long-term memory along with changes in the nucleus accumbens. ERK1 has a critical regulatory role in adaptive changes in the basal ganglia. This is in agreement with the observation that plasticity in the developing rat visual cortex involves ERK1 activation. Thus, ERK1 is implicated in the regulation of plasticity in the central nervous system. In addition, ERK1 activation has been implicated as an important mediator of neuronal death in neurodegenerative diseases, including corticobasal degeneration, through a caspase-3-independent mechanism. Therefore, abnormal activity or expression of the ERK1 protein may have a role in the pathogenesis of corticobasal degeneration.

Comments:
ERK1 appears to be a tumour requiring protein (TRP). The active form of the protein kinase normally acts to promote tumour cell proliferation. Abnormal functioning of the MAPK3 protein has been associated with several types of human cancer. In addition, over activation of the MAP kinase signalling pathway is involved in the malignant transformation of cancer cells, as demonstrated both in vivo and in vitro. Mutations resulting in the activation of the MAPK3 protein, but not necessary the over-expression of the MAPK3 protein, have been associated with tumorigenesis in several cancer types.
 
Gene Expression in Cancers:

TranscriptoNET (www.transcriptonet.ca) analysis with mRNA expression data retrieved from the National Center for Biotechnology Information's Gene Expression Omnibus (GEO) database, which was normalized against 60 abundantly and commonly found proteins, indicated altered expression for this protein kinase as shown here as the percent change from normal tissue controls (%CFC) as supported with the Student T-test in the following types of human cancers: Barrett's esophagus epithelial metaplasia (%CFC= -46, p<0.017); Cervical epithelial cancer (%CFC= +52, p<0.036); Malignant pleural mesotheliomas (MPM) tumours (%CFC= +101, p<0.03); Prostate cancer (%CFC= +116, p<0.011); Skin melanomas - malignant (%CFC= +52, p<0.023); T-cell prolymphocytic leukemia (%CFC= +109, p<0.072); and Uterine fibroids (%CFC= +70, p<0.036).
Mutagenesis Experiments:

Insertional mutagenesis studies in mice support a role for this protein kinase in mouse cancer oncogenesis.
Mutation Rate in All Cancers:

Percent mutation rates per 100 amino acids length in human cancers: 0.06 % in 24447 diverse cancer specimens. This rate is only -17 % lower than the average rate of 0.075 % calculated for human protein kinases in general.
Mutation Rate in Specific Cancers:

Highest percent mutation rates per 100 amino acids length in human cancers: 0.49 % in 805 skin cancers tested.
